Thalassemia and hemophilia are blood disorders, but that's the gist, yes. The trick with the X-Gene is that while it corresponds to a particular section of DNA, what it changes to is not consistent. So the fastest thing to do is probe for the non-mutant expression. Because that proves they're human.
The problem with just stopping there is that there's a ton of ways that stretch of DNA could be different, causing the probe to fail, some of which represent a mutation in the Gifted Youngster sense and some of which don't. I could list them out, but you've only got a few hours, so the cliff notes version is that you're looking for that section to be really weird.
